LKR10 and LKR13 KrasG12D-mutant LUAD cells (on a 129/Sv genetic background, previously generated in the laboratory of T. Jacks) were transiently transfected with Keap1 CRISPR–Cas9 KO plasmid (sc-424513-KO-2) or Stk11/Lkb1 CRISPR–Cas9 KO plasmid (sc-423192). Lentiviral vectors encoding Cre (expressed from PGK promoter) and an sgRNA (expressed from human U6 promotor) target known or putative tumor suppressors that are recurrently mutated in LUAD (or pan-carcinoa), such as Trp53.
